Kinetic measurements show that the organoruthenium radical [CpRu(CO)2]• prefers the sulfur atom, and the organomolybdenum radical [CpMo(CO)3]• prefers the selenium atom, in their reactions with diphenyl dichalcogenides.

A set of metal carbonyl cluster-boronic acid conjugates of the group VIII metals (Fe, Ru and Os) was synthesized and their antiproliferative effects measured against two breast cancers (MCF-7 and MDA-MB-231) as well as a non-cancerous breast epithelial (MCF-10A) cell line. The cytotoxicity followed the order Ru > Os > Fe for the MDA-MB-231 cells.

The tetraruthenium tetrahydrido carbonyl cluster Ru4(µ-H)4(CO)12 (1) reacts with diphenylvinylphosphine under thermal activation to give the substitution products Ru4(µ-H)4(CO)12-n(κ1-Ph2PCH=CH2)n, (2) (n = 3 (a); 4 (b)) in which each diphenylvinylphosphine ligand is on a different ruthenium metal center.

The diruthenium complex [CpRu(CO)2]2 reacts with diphenyl dichalcogenides via photoinitiation to [CpRu(CO)2]• radicals. The reaction with selenyl sulphide was slower than with disulphide. This has implications for the selective targeting of TrxR.

Ferrocenyl and ruthenocenyl derivatives of ebselen are found to be competitive inhibitors of mammalian TrxR, and are selective over GR and excess GSH. They exhibit lower Km values than ebselen and the natural substrate, Trx.

The osmium cluster 1 is the precursor to the metalloligand 2. Its TMNO-activated reaction with PPh3 in CH3CN leads to initial substitution at the Os(CO)3Cl2 “spike” which also shuts down release of the osmostibine. The CH3CN-substituted osmostibine could be generated in situ through the bis-acetonitrile derivative of 1 by converting the “spike” back into its removable Os(CO)3Cl2 form through reaction with CO gas, and then trapping it with W(CO)5(NCCH3).

The ruthenacyclic carbamoyl complexes [RuBr(2-NHC(O)C5H3N-4-R)(CO)2(MeCN)] in which the 4-position of the pyridinyl ring is derivatized, including by a phosphoryl group, have been synthesized. These bring the ruthenium-based structural mimics a step closer to the natural [Fe]-hydrogenase metal cofactor.

The solution-state isomers of the heteronuclear cluster RuOs3(µ-H)2(CO)12(PPh3) has been re-examined. The result is a reassignment of the structures and hence the exchange processes observed in the NMR spectra. The results affect the interpretations in at least three of our earlier papers!

Ruthenium-based structural mimics of the [Fe]-hydrogenase metal cofactor in which the acyl moiety is replaced by an N-heterocyclic carbene are reported. The pyridyl-NHC ligand has a rather different trans influence from that of a simple NHC.

The reaction of the triosmium carbonyl cluster Os3(CO)10(NCCH3)2 with a number of protected amino acids, tri- and pentapeptides show that reaction with the side chains follow the order: -SH > -COOH > -CONH2. There is a slight preference for the longer side chain of glutamic acid over aspartic acid. The trends observed are retained under physiological conditions and in the peptides.

The known cytotoxic and apoptosis-inducing osmium carbonyl cluster Os3(CO)10(NCCH3)2 is a non-substrate inhibitor of mammalian TrxR, with an IC50 of 5.3 ± 0.9 µM. Its inhibition of TrxR is selective over glutathione reductase and excess reduced glutathione. This inhibition has also been demonstrated in cell lysates, suggesting TrxR inhibition as a potential apoptotic pathway.

The higher nuclearity cluster [Os10(µ6-C)(CO)24]2- has been derivatized by reaction with SbX3 or p-tolylSbX2 via insertion of Sb into an Os-Os bond. Further insertion occurs with excess iodine. The chemistry and structure of these clusters can be understood in terms of the isolobal analogy between [Sb(p-tolyl)nX2-n)]+ and I+.

The complex [Ru2(CO)4(µ-O2CCH2OSi(OEt)3)2(PPh3)2] was used as a single-source precursor to prepare silica-encapsulated ruthenium. The material obtained at different stages of its hydrolysis catalyses different cycloaddition reactions between alkenes and ethyl diazoacetate.

The activated cluster Os3(CO)10(CH3CN)2 reacts readily with a number of carboxylated chalcones and benzaldehydes to afford products with the general formula Os3(CO)10(µ-H)(µ-O2CR).
